Empadronamiento is your registration on the population roll of your municipality, the padrón municipal, kept by the town hall (the ayuntamiento). The document that evidences it is called the certificado or volante de empadronamiento.
It's the equivalent of official proof of address: it shows where you actually live in Spain. Where other countries often make do with an electricity bill, Spain has a municipal register, and that's what counts.
Empadronamiento isn't just one more formality: it's the key that opens a good part of everyday life.

Spanish administrative procedures change, and practice varies from one province and one town hall to the next. In practice, the two are prepared in parallel. The NIE identifies you, empadronamiento gives you an address. Some town halls will register someone before they have their NIE, others won't; and conversely, proof of address makes other steps easier. This is exactly the kind of sequence you're better off not improvising, because every mistake in the order costs an appointment.

If you habitually live in a Spanish municipality, yes: registration on the padrón municipal is a legal obligation. It's also in your interest, because this document governs access to public healthcare, school enrolment and numerous other procedures.
Yes. A rental contract in your name is one of the most common supporting documents. If you're staying with someone, the town hall will generally accept written authorisation from the property holder along with their identity document and their own proof.
It's generally one of the quickest procedures: in many town halls the certificate is issued on the day of the appointment or within a few days. The real delay depends mostly on appointment availability in your municipality.
You need to register in your new municipality. Registering at the new town hall automatically removes you from the old one: there's no de-registration step for you to take.
